En mi experiencia realmente no es necesario cronometrar exactamente el tiempo con cristal violeta, iodo lugol y safranina debe de dejarse mínimo un minuto, pero realmente máximo tiempo no hay, y la decoloración no debe de ser hasta que deje de escurrir cristal violeta porque muchas veces y aunque se usen cepas jóvenes, se corre el riesgo de que se decolore a la fuerza, lo mejor es dejar derramar el chorrito de decolorante desde el inicio de la laminilla y permitir que llegue hasta el final de la misma, pasando por eso frotis ( que debe ser de buen tamaño y grosor) y de inmediato enjuagar. Crees ese es el "secreto" para una buena tinción.
